体外碱性成纤维细胞生长因子聚乳酸纳米缓释微球对人脂肪干细胞增殖和成脂分化的影响 被引量:3

Effect of basic fibroblast growth factor-polylactide sustained release nanuspheres on proliferation and adi- pollenic induction of adipnse-derived stem cells in vitro

摘要 目的体外脱察碱性成纤维细胞生长因子(hasic fibroblast growth factor,bFGF)聚乳酸纳米缓释微球对人脂肪干细胞增殖和成脂分化的影响,为bFGF缓释做球应用于脂肪组织工程的研究提供理论依据。方法体外分离培养脂肪干细胞,并行多向诱导分化鉴定。配制含有0、1、2、3、11、5mg/mlbFGF聚乳酸缓释微球的脂肪干细胞培养液及成脂分化诱导液。将脂肪下细胞按种至96孔板,第2灭更换含小同浓发hFGF缓释做球的培养液和成脂诱导液.分别用阴甲基偶氮噻哗蓝比色法(MTT)和油红O定量检测法定期检测细胞增雅和成脂分化的情况。所得数据均用SPSS13.0软件进行统计学处理。结果bFGF聚乳酸缓释微球行明显促进脂肪干细胞增殖和成脂分化的作用。增殖实验和成脂诱导实验合适的作用浓度分别为3mg/ml和4mg/ml。结论bFGF聚乳酸纳米缓释微球体外可以用显促进脂肪干细胞的增殖和成脂分化,可作为一种较理想的细胞因子缓释系统应用于脂肪组织工各国程的研究。 Objective To investigate the effeet of basic fibroblast growth factor polyactide re lease nanospheres on proliferation and adipogenic induction of adipose-derived stem cells in vilro. Methods Adipose-derived stem cells were isolated and induced for three-line differentiation in vitro. The cuhure medium and inductive medium of stem cells were prepared containing 0, 1, 2, 3, 4 and 5 mg/ml basic fibroblast growth factor polyaclide release nanosphercs, respectively. Adipose-derived slem eells were cuhured ina 96-well plate and replaced the cuhure medium containing release nanosph eres the second clay. The cells proliferation was detecled by the method of MTT every day and quanti- ficafion of oil red O every other clay. The data obtained were analyzed with SPSS13.0 statistical software. Results The basic fibroblas* grow',h faetor-polyactide release nanospheres bad the ability pro tooling proliferalion and adipogenic induction of adipose-derived stem cells. The best concentration of nanospheres was 3 mg/ml and 4 mg/ml, respectively. Conclusions The basic fibroblast growth fac for polyactide release nanospheres could promote proliferation and adipogenic inductionof adipose de rived slemcells significantly, h could be used as an ideal cytokine release nanospberes in adipose tissue engineering.
