摘要
目的:探讨化瘀消癥杀胚复方含药血清对体外培养的输卵管妊娠滋养细胞凋亡相关蛋白Bcl-2及Bax的影响。方法:取化瘀消癥杀胚复方含药血清及甲氨喋呤、生理盐水组血清对体外培养的输卵管妊娠滋养细胞进行培养,通过蛋白质印迹法(Western Blot)检测与含药血清作用72h后,滋养细胞中凋亡相关蛋白Bcl-2和Bax的表达。结果:滋养细胞与含药血清共同作用72h后,可降低Bcl-2蛋白表达量,与对照组比较,差异具有显著统计学意义(P<0.01)。而Bax蛋白表达量未见有明显变化,两组含药血清与对照组比较,差异均无统计学意义。结论:化瘀消癥杀胚复方可能通过下调抗凋亡蛋白Bcl-2的表达,降低Bcl-2/Bax比值,从而促进凋亡的途径起到治疗输卵管妊娠的作用。
Objective： To observe the effects of apoptosis related protein Bcl-2 and Bax on the human tubal gestational trophoblast cultured in vitro that treated by medicated serum which composed by Huayu Xiaozheng Shapei Compound.Method： The human tubal gestational trophoblast were cultured by three different medicated serum,which were composed by Chinese medicine,Methotrexate and normal saline.Then the cells were cultured for 72h to be checked the apoptosis related protein Bcl-2 and Bax by western blot.Results： After had been treated with different serum that contained different medicine in vitro for 72 hours,the expressions of Bcl-2 protein of the trophocytes descended.There is a significant difference between the normal saline group and Chinese medicine group（P0.01）.There is no difference between the group of Methotrexate and group of Chinese medicine.After had been treated with different serum that contained different medicine in vitro for 72 hours,the expressions of Bax protein of the trophocytes didn,t change obviously.There is no difference among the three groups.Conclusion： Down regulating the expressions of Bcl-2,down regulating the ratio of Bcl-2/Bax and inducing apoptosis may be one of the mechanisms of the effect of the Huayu Xiaozheng Shapei Compound.
