摘要
考察了珍贵橙色束丝放线菌SIPI-3-21生产安丝菌素P-3(AP-3)的发酵工艺。首先优化了摇瓶发酵培养基配方,在含葡萄糖20.0 g/L、玉米淀粉30.0 g/L、棉籽饼粉30.0 g/L、CaCl2 10.0 g/L和CaCO3 5.0 g/L的发酵培养基中发酵培养9 d,AP-3产量为123.2 mg/L。进一步在摇瓶中考察了前体异丁醇对AP-3合成的影响,结果表明,添加异丁醇可以显著提高AP-3的产量。当其添加浓度为54 mmol/L时,AP-3的产量为307.8 mg/L,同时发酵液中AP-3的含量从57.2%提高到86.5%。
The fermentation process for the production of ansamitocin P-3 (AP-3) by Actinosynnema pretiosum SIPI-3-21 was investigated. By optimization of fermentation medium, the yield of AP-3 reached 123.2 mg/L in the medium containing glucose 20.0 g/L, corn starch 30.0 g/L, cotton seed meal 30.0 g/L, CaC12 10.0 g/L and CaCO35.0 g/L, cultured for 9 days in a shake flask. The effect of isobutanol on AP-3 production was also investigated. The results showed that the titer of AP-3 was 307.8 mg/L after feeding 54 mmol/L isobutanol during the fermentation process, and the component of AP-3 in the fermentation broth was increased from 57.2 % to 86.5 %.
