慢病毒转染骨髓间充质干细胞在脊柱融合中的作用

Effects of bone marrow mesenchymal stem cells transfected with lentivirus in spinal fusion

摘要 目的评价慢病毒转染的骨髓间充质干细胞诱导大鼠脊柱融合的能力。方法构建携带骨形态发生蛋白2(BMP-2)的慢病毒并将其转染到骨髓间充质干细胞中,观察细胞的碱性磷酸酶染色和活性。并将转染的细胞移植到大鼠脊柱融合模型中,8周后处死大鼠,对其脊柱标本进行手触力学测试,Micro-CT扫描和组织学切片观察。结果慢病毒转染细胞的碱性磷酸酶染色明显好于未转染细胞(3.2±0.4比1.1±0.2,P〈0.05)。转染的细胞移植到大鼠体内8周后,X线、Micro—CT、手触力学测试和组织学切片均显示转染的大鼠脊柱标本全部达到骨性融合。而未转染的大鼠脊柱标本均未融合。结论慢病毒转染的骨髓间充质干细胞能够诱导脊柱融合,可作为基因载体工具用于脊柱融合。 Objective To assess the spinal fusion in rats induced by bone marrow mesenchymal stem cells (BMSC) transfeeted with lentivirus. Methods Alkaline phosphatase (ALP) staining and ALP activity were determined in BMSC transfected with lentivirus encoding BMP-2 that were implanted to rat model of spinal fusion. At week 8, all rats were sacrificed. Thereafter, measurements of manual mechanics, Micro-CT scanning and histological analysis were performed. Results ALP staining appeared more satisfactory in BMSC transfeeted with lentivirus as compared with untransfected cells(3.2±0.4 vs 1.1±0.2, P〈0.05). At week 8, spine bony fusion was achieved in all the transfected rats, as indicated by radiographic evaluation, Micro-CT and manual mechanics. In contrast, spinal fusion was absent in untransfected rats. Conclusion Lentivirus-transfected BMSC may achieve well-induced spinal fusion, and lentivirus vectors may offer the application of spinal fusion.
