氨溴索对吸入性肺损伤大鼠肺组织PBEF表达的影响及其抗炎作用 被引量:4

Anti-inflammatory roles of ambroxol in rats' gastric aspiration lung injury and activity of ambroxol on pre-B-cell colony-enhancing factor

摘要 目的探讨氨溴索对胃内容物吸人性肺损伤大鼠动物模型的抗炎保护作用。方法54只健康Sprague-Dawley雄性大鼠随机分为3组:对照组、损伤组、氨溴索组。观察各组大鼠动脉血氧分压(Pa02)、肺组织湿重/干重比(W/D)、髓过氧化物酶(myeloperoxidase,MPO)活性变化;ELISA法测定血清肿瘤坏死因子n(TNF-a)含量;Westernblot方法检测肺组织前B细胞克隆增强因子(pre-B-cellcolony-enhancing factor,PBEF)蛋白表达;光镜下观察肺组织结构变化。结果与对照组比较,损伤组各时相点Pa0。降低(P〈0.05),肺组织W/D以及MPO活性增加(P〈O.05),血浆中TNF-a含量增高(P〈i0.05);PBEF蛋白表达均增高(P〈O.05);肺组织出现明显病理组织学损伤。与损伤组比较,氨溴索组各时相点Pa02升高(Pd0.05),肺组织W/O以及MPO活性降低(P〈0.05),TNF-a含量降低(Pd0.05);PBEF蛋白表达均下降(尸d0.05);肺组织病理学损伤减轻。结论在大鼠胃内容物吸人性肺损伤模型中,氨溴索可通过抑制肺组织PBEF表达、抑制炎症介质的释放发挥抗炎保护作用。 Objective To investigate the anti-inflammatory roles of ambroxol in rats' gastric aspiration lung injury. Methods Fifty-four Sprague-Dawley rats were randomly divided into three equal groups ~ control group, injury group and ambroxol group. Arterial partial pressure of oxygen (PaO2), wet- to-dry weight ratio (W/D) of lung, the activity of myeloperoxidase (MPO) were measured. Tumor necrosis factor-a (TNF-a) of serum were examined by enzyme linked immunosorbent assay. Pre-B-cell colony-enhancing factor (PBEF) was examined by Western blot. Lung pathological change was observed by microscope. Results Compared with control group, PaO2 decreased but W/D of lung,the activity of MPO,TNF-a of serum and PBEF expression in injury group increased (all P d0.05). Compared with injury group, PaO2 increased but W/D of lung, the activity of MPO, TNF-a of serum and PBEF expression in ambroxol group decreased (all P d0.05). The microscopic structure of the control group was normal and the pathological changes of ambroxol group were milder than those of injury group. Conclusions Ambroxol has anti-inflammatory effects by restraining the activity of PBEF and the releasing of inflammatory cytokines on gastric aspiration lung injury.
