摘要 目的了解诺卡菌的培养及染色特征。方法采用脑心浸液琼脂培养基、哥伦比亚血琼脂基础培养基和沙保弱琼脂培养基,分别在25℃和37℃条件下培养,观察不同种类诺卡菌的生长时间和菌落生长形态。应用革兰染色、抗酸染色及改良抗酸染色方法进行染色,观察诺卡菌的染色特征和形态特点。结果共对从德国的DSMZ菌种保藏中心购得的7个种23株诺卡菌标准参照菌株进行了培养观察。诺卡菌的菌落形态和颜色多样,菌落形态有针尖状、雪花状、菜花状、不规则状等,菌落颜色有白色、黄色、橙红色、灰色等。诺卡菌为革兰氏阳性菌,弱抗酸性菌,用不同浓度的脱色液进行抗酸染色,所用脱色液浓度越低,抗酸染色阳性率越高,硫酸脱色液浓度为0.125%时,诺卡菌抗酸染色阳性率最高。结论诺卡菌具有独特的培养和染色特征,菌落形态和镜下形态多样。诺卡菌为弱抗酸菌,0.125%硫酸脱色液的改良抗酸染色方法检测阳性率最高。 To investigate the cultivation and staining characteristics of Nocardia,3 different media including Brain Heart Infusion Agar,Columbia Blood Agar Base and Sabouraud Dextrose Agar were used for incubating 23 Nocardia reference strains.The growth status,time and colony morphology of Nocardia at 25℃ and 37℃ were observed.The culture features and stain characteristics were observed by means of Gram's stain,Kinyoun acid-fast stain,and modified Kinyoun acid-fast methods.Total of 7 species including 23 strains from DSMZ(Germany) were observed.Colonial morphology and colour of Nocardia were variable.The colonial morphology was needle-shaped,snow-shaped,cauliflower-shaped and irregular-shaped appearance.Nocardia colonies varied from white to yellow,orange and grey in colour.Nocardia species were gram positive and weakly acid-fast bacteria.The higher the positive rate of modified acid-fast staining,the lower the concentration of decolourizer,when using different concentration of sulfuric acid as decolourizer.The positive rate was the highest,when using 0.125% sulfuric acid for decolourization.Nocardia species have special cultivation features and unusal staining characteristics.
