摘要
目的:探讨乳腺浸润性导管癌组织中p27、Skp2和E-cad的表达及其生物学意义。方法:采用微阵分析免疫组化法检测p27、Skp2和E-cad在浸润性导管癌组织、淋巴结转移灶与乳腺良性病变中的表达强度。结果:浸润性导管癌组织中p27和E-cad的表达强度低于乳腺良性病变,χ2分别为13.648、63.163,P值均<0.05;Skp2的表达强度高于乳腺良性病变,χ2=17.150,P<0.05。在浸润性导管癌组织中Skp2+/p27低指数与核异形性、组织学分级和癌巢分化程度及淋巴结转移情况具有相关性,χ2分别为12.634、6.397、8.750、23.768,P值均<0.05,与间质纤维化程度、年龄和肿物大小无相关性,χ2分别为1.325、0.281、3.748,P值均>0.05。浸润性导管癌淋巴结转移灶的E-cad表达强度低于原发瘤,χ2=23.142,P<0.05。结论:p27、Skp2和E-cad在乳腺肿瘤中与肿瘤分化程度、组织学分级和肿瘤转移有关。Skp2+/p27低指数和E-cad表达强度可能对乳腺癌患者的诊断及预后评估有意义。
OBJECTIVE: To investigate the expression of p27,Skp2 and E-cad in invasive ductal carcinoma(IDC),and their biological significance.METHODS:The protein expression of p27,Skp2 and E-cad,was detected in microarraied tissue chip of primary tumor(PT),and lymphonodi metastatic tumor(LMT),of invasive ductal carcinoma(IDC),by using immunohistochemical method and benign proliferature lesions(fibroadenosis and fibroadenoma of breast) as controls.RESULTS:The expressing intensity of p27 and E-cad was much higher in benign lesions than that in IDC tissues(χ2 were 13.648 and 63.163,P0.05).The expression of Skp2 in malignancies was higher than that in benign lesions(χ2 =17.150,P0.05).The index of Skp2+/p27low was correlated with nuclear atypia of tumor cells and size of cancer nest and grade of histology and metastasis(χ2 were 12.634,6.397,8.750 and 23.768,P0.05),and was not correlated with interstitial fibrosis,age and tumor size(χ2 were 1.325,0.281 and 3.748,P0.05).The expressing intensity of E-cad was much lower in IDC-LMT than that in IDC-PT(χ2=23.142,P0.05).CONCLUSION:The expressing level of p27,Skp2 and E-cad are related to the differentiation degree and metastatic potential of breast carcinoma,and the Skp2+/p27low index and expression of E-cad may be used to evaluate the prognosis of patients with breast carcinoma.
