督脉电针刺激对脊髓全横断小鼠大脑皮质运动区BDNF表达的影响 被引量:9

Effects of Electroacupuncture of "Governor Vessel" Acupoints on Changes of BDNF in the Cortical Motor Area of Mice with Spinal Cord Transection

摘要 目的研究督脉电针刺激对脊髓全横断(spinal cord transection,SCT)小鼠大脑皮质运动区脑源性神经营养因子(brain derived neurotrophic factor,BDNF)表达的影响。方法将绿色荧光蛋白转基因小鼠在T9、T10间行SCT,随机分为电针(EA)组、对照组,每组9只小鼠。EA组于术后第1d开始电针刺激至第13d结束,对照组仅做SCT。术后28d时,取小鼠大脑皮质运动区脑组织,用免疫组化、原位杂交、逆转录-聚合酶链式反应、酶联免疫吸附试验检测皮质运动区BDNF的表达变化。结果 BDNF蛋白和mRNA阳性产物主要表达于皮质运动区神经元。EA组、对照组皮质运动区BDNF蛋白表达分别为(1973.41±194.71)pg/mg、(1615.22±137.21)pg/mg,差异有统计学意义(P<0.05);而BDNFmRNA表达水平差异无统计学意义(P>0.05)。结论脊髓损伤后督脉电针刺激能够有效增加小鼠大脑皮质运动区BDNF蛋白表达,这有助于揭示电针刺激促进脊髓损伤修复的机制。 Objective To investigate effects of electroacupuncture of "Governor Vessel" acupoints on changes of brain erived neurotrophic facotr(BDNF) in the cortex area of mice with spinal cord transection(SCT).Methods GFP mices were made spinal cord transection between T_9 and T_10,and then divided into electroacupuncture group(EA group) and control group;mices of EA group had be given EA from 1 day postoperation to 14 day postoperation,however,mices of control group had only SCT.Immunohistochemistry,in situ hybridization,RT-PCR and ELISA were performed to observe changes of BDNF in the cortical motor area of EA and control group.Results At 28 d postoperation the protein expression of BDNF in the cortex area of EA group and control group was(1973.41±194.71) pg/kg and(1615.22±137.21) pg/kg respectively,and there was statistical difference between them(P0.05).However,there was no obvious different in expression of BDNF mRNA in the cortex area between EA and control group(P0.05).The positive products of BDNF mRNA and protein were mainly located in neurons in the cortex area.Conclusion EA in "Governor Vessel" can effectively induce the increases of BDNF protein in the cortex area which may be helpful to understand the mechanism of EA in the treatment of spinal cord injury.
