摘要
目的:观察中药复方JEYS(TCCJEYS)对大鼠离体小肠平滑肌的解痉作用及作用机制。方法:制备Wistar大鼠离体小肠平滑肌,观察TCCJEYS对乙酰胆碱(ACh)、氯化钙(CaCl2)和氯化钾(KCl)所致平滑肌收缩的影响。改变营养液中钙含量或用酚妥拉明和普萘洛尔分别对标本进行预处理,观察TCCJEYS对ACh所致小肠平滑肌收缩作用的影响。结果:与对照组比较,TCCJEYS能明显抑制ACh、CaCl2和KCl引起的Wistar大鼠离体小肠平滑肌收缩(P<0.05或P<0.01),且对内钙外流和外钙内流均有显著抑制作用(P<0.01);预先给予酚妥拉明或普萘洛尔后,TCCJEYS对离体小肠平滑肌的舒张效应没有变化(P>0.05)。结论:TCCJEYS对大鼠离体小肠平滑肌具有解痉作用,且其作用与非选择性抑制钙通道有关。
Objective: To observe spasmolysis effects of chinese herb compound JEYS on isolated small intestine smooth muscle of rats,and to explore its mechanism.Methods: Small intestine smooth muscle of wistar rats was isolated for the observation of JEYS effects on smooth muscle contraction evocated by acetylcholine(Ach),CaCl2 and KCl,and the contraction evocated by ACh after changing Ca2+ concentration or pretreating samples with phentolamine and propranolol.Results: Compared with the control group,JEYS significantly inhibited contraction of isolated small intestine muscle evocated by ACh,CaCl2 and KCl(P0.05 or P0.01),and significantly inhibited outflow of intracellular Ca2+ and inflow of extracellular Ca2+(P0.01).After pretreating samples with phentolamine or propranolol,relaxation effects of JEYS on the isolated small intestine smooth muscle didn't change(P0.05).Conclusions: JEYS has spasmolysis effects on isolated small intestine smooth muscles of rats,and the effects relate to non-selective inhibition of calcium channel.
