摘要
目的探讨PIAS3过表达对人脑胶质瘤U251细胞生长的作用及其可能的机制。方法通过实时荧光定量PCR检测PIAS3在正常脑细胞和脑胶质瘤U25 1细胞中的表达情况。转染PIAS3过表达载体,提高U251细胞中PIAS3表达水平,通过噻唑蓝(MTT)试验检测细胞增殖情况。Western Blot验证PIAS3表达与增殖相关基因PI3K及Akt间关系。结果 PIAS3在正常脑组织中相对高表达,而在人脑胶质瘤U251细胞中表达明显下降(P<0.01)。体外转染PIAS3过表达载体能明显抑制U251细胞生长,并且有效抑制PI3K活性及p-Akt表达,但对总的Akt无明显影响。结论胶质瘤U251细胞中异常低表达的PIAS3发挥着重要的增殖促进作用,过表达PIAS3可通过抑制PI3K/Akt通路有效抑制U251细胞增殖。
Objective To investigate the function and mechanism of protein inhibitor of activated signal transducer and activator of transcription 3 (PIAS3)overexpression on the growth of U251 cells. Methods The expression of PIAS3 on normal brain tissue and human glioma U251 cells was quantified by real-time fluorescence quantitative RT-PCR. U251 cells were transfected with PIAS3 overexpression vectors to improve the level of PIAS3 expression in U251 cells and then the cell pro- liferation was measured by MTT assay. The relationship between PIAS3 expression and proliferationrelated genes PI3K and Akt was identified by Western blot. Results PIAS3 was relatively highly expressied in normal brain tissue ,but obviously downregnlated in U251 cells(P 〈 0.01 ). And our results showed transfection with PIAS3 overexpression not only remarkably induced proliferation inhibition of U251 cells,but also inhibited the PI3K activity and p-Akt expression. However,the expression of total Akt was not affected. Conclusions The low-expression of PIAS3 functions an important role on the state of proliferation in U251 cells, and upregulation the expression of PIAS3 can effectively inhibit the proliferation of U251 cells through inhibiting the activation of PI3K/Akt pathway.
