摘要
目的观察全天麻胶囊对血管性痴呆大鼠认知能力及额叶Bcl-2、Bax蛋白表达的影响,探讨全天麻胶囊的神经保护作用。方法采用双侧颈总动脉永久结扎法制备血管性痴呆动物模型,全天麻胶囊高低剂量组分别给予灌服全天麻胶囊混悬液10 g(kg.d)及5g/(kg.d),对照组均给予等量生理盐水灌胃。以Morris水迷宫方法检测大鼠认知能力;以免疫组化方法检测大鼠额叶Bcl-2、Bax蛋白的表达。结果与对照组相比,模型组认知能力明显下降(P<0.05),Bcl-2、Bax蛋白表达增加(P<0.05);与模型组相比,全天麻高低剂量组认知能力明显改善(P<0.05),Bcl-2蛋白表达明显增加(P<0.05),Bax蛋白表达明显减少(P<0.05),其中高剂量组较低剂量组效果更明显(P<0.05)。结论血管性痴呆大鼠给予全天麻胶囊后,可以通过调节额叶Bcl-2及Bax蛋白的表达,抑制神经细胞凋亡,促进脑功能的恢复,从而改善大鼠学习记忆能力。
Objective It is to observe the influence of Quantianma capsule on cognitions and the expression of Bcl - 2 and Bax protein in frontal lobe of vascular dementia (VD) rat, and approach its' neurology protective effect. Methods Rat models of VD were made by adopting the permanentocclusion of bilateral carotid arteries in Wistar rats. Morris water maze was used to test the cognitive function of these rats, the expression of Bcl - 2 and Bax protein in the frontal lobe were detected with immunohistochemical technique. Results Compared with control group, cognitions was remarkably decreased and the expression of Bcl - 2 and Bax protein in frontal lobe were increased in VD model rat ( all P 〈 0.05 ). Compared with model group, cogni- tions and the expression of Bcl -2 protein was increased and expression of Bax protein was reduced in Quantianma capsule groups ( all P 〈 0.05 ) ; the expression of Bcl - 2 protein in high-dose group was increased than that in low-dose group and the expression of Bax protein in high-dose group was reduced than that in low-dose group ( all P 〈 0.05) . Conclusion Quantianma capsule can inhibit apoptosis through regulating the expression of Bcl - 2 and Bax protein in the frontal lobe. Quantianma capsule may improve cognitions in VD rats.
