摘要
近年来,随着免疫学研究的不断深入,疫苗研制技术取得了很大的进步,但这些疫苗存在着给药剂量大、免疫原性弱和靶向性差等缺点,需加入免疫佐剂以提高其免疫原性和特异性免疫应答。纳米疫苗黏膜免疫递送系统具有保护抗原、简化接种程序、增强免疫效果和提高生物利用度等优点。壳聚糖及其衍生物由于具有良好的可生物降解性、生物相容性、低毒性、黏膜吸附性、免疫刺激活性、缓释和控释作用及靶向性,已成为了构建纳米疫苗黏膜免疫递送系统和疫苗免疫佐剂的优良载体。文中对疫苗递送系统、壳聚糖及其衍生物的理化性质及其作为免疫佐剂的应用、优越性和作用机制进行了综述。
In recent years,great progress in the preparation of vaccine has been achieved with the deep research of immunological technology.However,these vaccines have disadvantages such as low effects(requiring large dose),weak immunogenicity,unsatisfactory targeting properties.Thus immune adjuvants are required.The mucous immune processing system based on nanovaccine can protect antigen,simplify vaccination procedure,enhance vaccination effects,and improve bioavailabilities.Chitosan and its derivatives have become a kind of greatly potential adjuvant for vaccine immune due to its good biocompatibility and biodegradability,low toxicity,mucosa adsorption and immune stimulating activity,sustained and controlled release profiles,and targeting effects.In this paper,we reviewed the vaccine delivery system,physico-chemical properties and application of chitosan and its derivatives as the immune adjuvant.The superiority and mechanisms of these systems were also elaborated.
