TGF-α在B6-Co小鼠胚胎发育后期眼睑中表达变化 被引量:4

Changes of TGF-α expression in eyelids of B6-Co mice during later stage of embryonic development

摘要 目的检测转化生长因子α(TGF-α)在遗传性角膜混浊(B6-Co)小鼠胚胎发育后期眼睑中的表达变化。方法雄性B6-Co和B6小鼠分别与雌性B6小鼠以1∶2比例配对,各10笼。体视显微镜下取不同胚胎期小鼠眼睑组织,采用实时定量PCR法和免疫组化法分别检测眼睑组织中TGF-αmRNA和蛋白表达。结果随着发育时间的推移,B6-Co小鼠上、下眼睑不能融合,角膜完全暴露;TGF-αmRNA表达低于B6小鼠(P<0.05),而TGF-α蛋白表达与B6小鼠比较差异无统计学意义。结论 TGF-α表达可能是导致B6-Co小鼠胚胎期眼睑发育缺陷的主要原因之一。 Objective To investigate the changes of transforming growth factor-α (TGF-α) expression in the eyelids of C57BL/6-eornea opacity(B6-Co) mice during later stage of embryonic development. Methods Both male B6-Co mice and male B6 mice were paired with female B6 mice at a ratio of one to two in ten cages eacK The eyelid tissues of mice during different stages of embryonic development were taken under stereomicroscope, and the expressions of TGF-α mRNA and protein were detected by RT-PCR and immunohistochemistry, respectively. Results As development time went on, the upper eyelid and lower eyelid were not closed and the cornea was completely exposured in B6-Co mice. Moreover,the expression of TGF-α mRNA in B6-Co mice was lower than that in 136 mice (P〈0.05), but there was no significant difference of expression of TGF-α protein between B6-Co mice and 136 mice. Conclusion TGF-α expression may be one of the main factors leading to abnormal eyelid development in B6-Co mice.
