放疗结合电化疗对小鼠肿瘤组织细胞G1期阻断与凋亡的诱导 被引量:5

G1 arrest and apoptosis of mouse tumor after radiotherapy and electrochemical therapy

摘要 目的 研究放疗结合电化疗对小鼠肿瘤组织的抑制作用。方法 将小鼠随机分为未治疗对照组、放射治疗组、电化学治疗组及放疗结合电化疗组,种植小鼠骨肉瘤细胞株S180,采用比较肿瘤生长曲线、流式细胞光度术及电泳等方法,进行多方面的分析。结果 与未治疗组相比,3个治疗组中肿瘤生长均得到有效抑制,在治疗后8h,对照组中G1期细胞只有41.5%,而单放疗,单电化疗及结合组G1期分别占70.3%,72.8%,78%。在16h未治疗对照组中没有凋亡细胞出现,而3个治疗组中凋亡细胞分别占13.0%、18.8%、31.3%,电泳也显示有明显的DNA连续性降解片断。结论 肿瘤生长曲线表明两种方法的结合,有协同抑制作用。流式细胞光度术及电泳检测结果表明,放疗或电化疗均能引起细胞G1期阻滞,诱导细胞出现凋亡,而两者结合也能将细胞强烈阻滞于G1期,并能进一步诱导细胞凋亡。这些结果显示放疗与电化疗间有协同抗癌作用,其诱导细胞引起G1期阻滞及凋亡之间可能有密切的相关性。 Objective To analyze the cooperative anti tumor effects of radiotherapy(RT) and electrochemical therapy(ECT) in mouse.Methods Mice of Kunming transplanted S 180 ascites carcinoma were used throughout this study.The experimental mice were divided into four groups:A,untreated control,B,RT alone,C,ECT alone,and D,RT plus ECT.The methods of tumor growth curve,flow cytometry and electrophoresis were used for analysis.Results The tumor growth curve showed that the tumor growth was delayed in group B,C,D especially in D.Using FCM and electrophoresis,the results showed that in 8 h the percentage of G1 phase in group B,C and D were respectively 70.3 %, 72.8 %,78 %,compared to 41.5 % in A,and in 16 h the percentage of apoptotic cells in group B,C and D,were 13.0 %, 18.8 % and 31.3 %,compared to 0 % in A.Obvious DNA ladders were presented in electrophoresis in the three treated groups at this time.Conclusion This study suggested that RT and ECT have the cooperative anti tumor effects.This study also revealed there were marked G1 arrest and increased apoptosis promoting cell death when combining radiotherapy with electrochemical therapy.Those indicated that the co operative anti tumor effect between RT and ECT is related closely to the induce of G1 arrest and apoptosis in mice tumor cells.
