摘要 研究牛microRNA的进化模式和现状,阐明microRNA的功能性SNP的作用和对动物表型的影响。选取牛per-microRNA及其相邻区域,统计各区域SNP数量和密度,考察SNP对microRNA二级结构稳定性和靶基因选择的影响。结果显示,per-microRNA的SNP密度显著低于其侧翼区,Seed区的SNP的密度较高,Seed区的SNP变异对per-microRNA二级结构稳定性的影响最高,microRNA成熟体SNP可能影响mi-croRNA对靶基因的选择。上述结果表明,牛在进化过程中可能存在加速进化的现象,目前牛microRNA的进化正处于活跃期。microRNA上的功能性SNP位点对microRNA的靶基因选择和结构稳定性有很大影响,并最终通过microRNA来影响动物表型的改变。 This study aims to study the evolution of cattle microRNA patterns and the status quo,to clarify the functional SNP on microRNA's role and influence of animal phenotype.Select cattle per-microRNA and its neighboring regions,statistics of the volume and density SNP regions to study SNP stability and secondary structure of microRNA target gene of choice.The SNP density in pre-microRNA regions was significantly lower in the flanking regions;seed zones of higher density SNP;the SNPs in the Seed regions showed greater effects on the stability of the pre-microRNA secondary structure than those SNPs in other regions did;the SNPs in the mature microRNA regions also appeared to influence target selection of the microRNA.These results indicate that cattle may exist in the evolutionary process to accelerate the evolution of the phenomenon,the evolution of the current cattle microRNA are active period;microRNA on the functional SNP sites of the microRNA target gene selection and the structure has great influence on the stability,ultimately,to influence the animal through the microRNA phenotypic changes.
