摘要
目的检测大鼠精原干细胞早期发育过程中第6天与第8天睾丸组织的差异表达基因。方法通过基因芯片技术,利用Roche.Nimble Gen公司大鼠12×135K全基因组表达谱芯片分别与第6天和第8天睾丸组织的cDNA进行杂交。比较两个样品差异表达的基因。结果差异表达基因700个,表达上调的基因共295个,表达下调的基因共405个。参与了1407个基因功能分析条目;参与了46个生物学通路。结论大鼠精原干细胞早期发育过程中,精原干细胞的发生与增殖是一个多基因参与、多因素作用和多阶段进展的过程。
Objective To detect the testicular gene differential expression on the 6^th and 8^th days in rat's prepuberty in the developmental period of rat' s spermatonial sperm stem cells. Methods Individual rat testes were collected on the 6^th and 8^th days. Total RNA was extracted and cDNA hybridized with the NimbleGen Hybridization System by gene chip technology. Results The microarray results showed that 700 genes, covering 1 407 GO terms and 46 signaling pathways, were differentially expressed between the rats of the 6^th day and the 8^th day. Conclusion In the early development of rat's spermatonial stem cells, their occurrence and proliferation are the result of the joint working of multiple genes and many other factors in a process of several stages.
