金纳多对大鼠离体肺缺血再灌注时肺组织中水通道蛋白1表达的影响 被引量:6

Effect of Ginaton on expression of aquaporin-1 in rat lung with ischemia-reperfusion

摘要 目的观察金纳多预处理后,水通道蛋白-1(AQP-1)在大鼠离体肺缺血再灌注损伤过程中的表达及其功能。方法将40只大鼠随机分为供体组和受体组(rt=20)。供体组再随机分为2组(n=lo),低钾右旋糖苷液(LPD)灌注组与LPD+金纳多组;受体组也随机分为2组(n=10)。LPD灌注组大鼠左肺设为A组,右肺设为B组,为对照组。LPD+金纳多灌注组左肺设为C组,右肺设为D组,为对照组。建立大鼠离体肺缺血再灌注模型,分组造模后,取肺组织,免疫组织化学法检测肺组织AQP-1的表达;RT-PCR检测肺组织AQP-1mRNA的表达;免疫印迹法检测肺AQP-1蛋白的表达。结果A组及c组肺组织中AQP-1蛋白表达的相对灰度值分别为0.65±0.06、0.88±0.11,两者比较差异有统计学意义(P〈0.05)。AQP.1在A组及C组肺组织中的mRNA表达平均相对A值分别为0.30.4-0.08、0.49±0.11,两者比较差异有统计学意义(P〈0.05)。结论肺移植术前给予金纳多治疗,可减轻供肺的损伤。 Objective To explore the expression and function of aquaporin-1 ( AQP-1 ) in rat lung with ischemia/reperfusion (I/R) injury after pretreatment with Ginaton. Methods Adult Wistar rats were divided into 2 groups. Twenty rats served as donors ( n = 20), and 20 as recipiems ( n = 20). Left lungs of donor rats were used as isolated lung reperfusion model and perfused only with LPD solution as group A (n = 10). The rats pretreated with Ginaton before reperfusion served as group C (n = 10). Right lungs of donor rats without any treatment served as control group ( group B and group D, n = 10 each). After the model was established, the expression of AQP-1 in the lung tissues was detected by using immunohisto- chemistry, Western blotting, and reverse transcriptase-polymerase chain reaction. Results Immunohisto- chemical examination revealed that AQP-1 was expressed in endothelia. Immunoblotting studies demonstra- ted that AQP-1 protein in groups A ant C was 0. 65 + 0. 06 and 0. 88 + 0. 11 respectively. The mRNA ex- pression in groups A and was 0. 30 +0. 08 and 0. 49 +0. 11 respectively. The expression of AQP-1 protein and mRNA in group C was significantly higher than in group A ( P 〈 0. 05 ). Conclusion The pretreatment with Ginaton can reduce the acute lung injury caused by I/R injury.
