摘要
目的探讨胞质磷脂酶A2(cPLA2)在肺癌中的表达及生物学意义,为探讨肺癌发生和发展的分子机制提供理论依据。方法采用反转录一聚合酶链反应(RT—PCR)技术检测60例肺癌及癌旁组织中cPLA2mRNA的表达,同时采用免疫组织化学sP法检测相应标本中cPLA2蛋白的表达。结果cPLA2mRNA在肺癌及癌旁组织中的相对表达量分别为0.82±0.10和0.08±0.01,与癌旁组织比较,肺癌的cPLA2的表达量明显增加(t=71.868,P=0.001)。肺癌组织中cPLA2蛋白阳性表达率为76.7%(46/60),癌旁组织未见表达。肺癌cPLA2蛋白的阳性表达率与肿瘤大小无关(χ2=0.366,P〉0.05);随着组织类型、淋巴结的转移、组织分化程度的降低和临床分期的增加,肺癌cPLA2蛋白的阳性表达率逐渐增高,差异有统计学意义(χ2值分别为7.565、4.424、9.161、5.416,均P〈0.05)。结论cPLA2的高表达可能在肺癌的发生、发展起着重要作用,可能为肺癌的早期诊断和开发肺癌的靶向治疗提供一定的临床依据。
Objective To investigate the expression and biological significance of cytosolic phospholipase A2 (cPLA2) in lung cancer and to provide a theoretical basis for understanding the molecular mechanism of the lung cancer carcinogenesis and development. Methods The expression of ePLA2 at mRNA level in tumor and tumor-adjacent tissue was detected by RT-PCR. The immunohistochemical SP method was used to evaluate the expression of cPLA2 protein in the corresponding tissues. Results The quantitive expression levels of cPLA2 mRNA in tumor and tumor-adjacent tissue were 0.82±0.10 and 0.08± 0.01, respectively. The mRNA level of cPLA2 significantly increased (P =0.001) in lung tumor when compared with those in the tumor-adjacent tissue. By immunohistochemistry, the positive rate of cPLA2 protein expression in lung tumor tissue was 76.7 % (46/60), while no expression was detected in tumor-adjacent tissue. The positive rate of cPLA2 protein expression in lung cancer was correlated with clinicopathological parameters including histological type, histological grade, lymph node metastasis and TNM staging (χ2 was 7.565, 9.161, 4.424, 5.416, respectively, and all P 〈0.05), but not correlated with the size of tumor (χ2 = 0.366, P 〉 0.05). Conclusion The expression of cPLA2 in lung cancer may play an important role in the process of carcinogenesis, and may provide a clinical basis for the early diagnosis and targeted therapy for lung cancer.
