摘要
目的:制备黄药子凝胶并建立其含量测定方法。方法:采用卡波姆为凝胶基质制备黄药子凝胶;采用高效液相色谱法测定黄药子中活性成分薯蓣皂苷元的含量,色谱柱为Alltech C18,流动相为乙腈-水=75∶25,流速为1 mL.min-1,柱温为30℃,检测波长为303 nm,进样量为10μL。结果:本凝胶质地均匀细腻,容易涂展,黏稠度适宜;薯蓣皂苷元检测浓度线性范围为0.08~0.8mg.mL-(1r=0.999 6),平均加样回收率为98.79%(RSD=0.88%,n=5)。结论:该制剂制备工艺简单,含量测定方法简单、准确、可靠。
OBJECTIVE: To prepare Dioscorea bulbifera gel and to establish the method for the content determination. METH- ODS: The Dioscorea bulbifera gel was prepared using carbopol as gel matrix . The content of active component diosgenin in Dioscorea bulbifera gel was determined by HPLC. The determination was performed on Alltech C18 colunin with mobile phase consisted of acetonitrile-water=75 : 25 at the flow rate of 1 mL.min-L The column temperature was 30 and detection wavelength was set at 303 nm. Injection volume was 10 μL. RESULTS: Dioscorea bulbifera gel was even and delicate, easily painted, suitable viscosity. The linear range of diosgenin was 0.08-0.8 mg-mL- (r=0.999 6) with an average recovery of 98.79% (RSD=0.88%, n = 5). CONCLUSION: The preparation technology is simple, and the method of content determination is simple, accurate and reliable.
