黄芪多糖对环磷酰胺引起小鼠骨髓抑制后造血功能的修复作用 被引量:17

摘要 目的探究黄芪多糖对环磷酰胺诱导的骨髓抑制小鼠骨髓造血功能的修复作用。方法腹腔注射150mg/kg/d环磷酰胺3d,制备小鼠骨髓抑制模型。实验分对照组、模型组、黄芪多糖高(100 mg/kg/d)、低(50mg/kg/d)剂量组。黄芪多糖腹腔注射给药7d,眶静脉取血测外周血血常规,取右侧股骨计数骨髓细胞数,并作股骨切片进行组织病理学检查。结果①与对照组比较,模型组骨髓细胞数明显减少;与模型组比较,黄芪多糖高剂量组骨髓细胞数显著增高,且明显改善骨髓腔组织的形态学变化;②外周血的血常规结果显示,模型组的红细胞、白细胞、淋巴细胞、单核细胞及血小板与正常组比较均被明显抑制,而给予黄芪多糖的高、低剂量组不同程度的改善了外周血象。结论黄芪多糖对环磷酰胺诱导骨髓造血功能抑制有修复作用。 Objective To study the restorative effect of Astragalus Polysaccharides(APS) on hematopoietic function in cyclophosphamide-induced bone marrow(BM) depression in mice.Methods Mice were injected by peritoneal injection(i.p.) with 150 mg/kg cyclophosphamide for three days for establishing BM depression model.Animals were divided into four groups,which were control group(C),model group(M),high dose group(H,100 mg/kg/d) and low dose group(L,50 mg/kg/d).After APS was administrated by i.p.for 7 days,the blood from orbital venous was used for peripheral blood routine.The right femur was taken for counting bone marrow cells,and the histopathology of femoral bone sections were examined by H.E.staining.Results ①Compared with the control group,BM cells was significantly reduced in model group;Compared with model group,BM cells were significantly increased and BM organization morphological changes were significantly attenuated after treatment with APS 100 mg/kg/d.②The results from blood routine showed that red cells,white cells,lymphocytes,monocytes,and platelets in model group were significantly decreased compared with control group.However,which were improved to a certain extent after APS at doses of 100 and 50 mg/kg/d.Conclusions APS may restore hematopoietic function in cyclophosphamide-induced bone marrow depression in mice.
