摘要
目的:利用新生大鼠缺血缺氧脑损伤动物模型,检测MMP-9的表达,探讨丙酮酸乙酯对MMP-9表达的影响及对缺血缺氧性脑损伤的保护作用。方法:7日龄清洁级Wistar大鼠90只随机分为三组:阴性对照组;缺血缺氧组:腹腔注射生理盐水1.5mL;丙酮酸乙酯治疗组:腹腔注射丙酮酸乙酯溶液50mg/kg。HE染色观察缺血缺氧后大鼠脑组织病理改变,免疫组化法检测MMP-9的表达。结果:HE染色见阴性对照组脑结构完整细胞形态正常;缺血缺氧组脑内见液化坏死灶,细胞肿胀排列紊乱,胞质淡染,核固缩裂解;丙酮酸乙酯治疗组改变较缺血缺氧组减轻。免疫组化结果显示,缺血缺氧组和丙酮酸乙酯治疗组MMP-9阳性细胞数明显多于阴性对照组,差异有统计学意义(P<0.01);丙酮酸乙酯治疗组MMP-9阳性细胞数较缺血缺氧组明显减少差异有统计学意义(P<0.01)。结论:缺血缺氧时新生大鼠脑组织中MMP-9表达增加;丙酮酸乙酯可以降低MMP-9的表达,具有脑保护作用。
Objective: To observe the change of MMP-9 expression after hypoxic-ischemic brain damage (HIBD) in neonatal rat models and to evaluate the protective role of ethyl pyruvate(EP) in hypoxic-ischemic brain injury. Methods:aged 7-days, ninety Wistar rats were randomly divided into three groups: negative control group;HIBD group: intraperitoneal injection of saline 1.5mL; EP group: intraperitoneal injection of ethyl pyruvate solution 50mg/kg. Rat brain pathology after hypoxic--ischemic was observed by HE staining and, MMP-9 expression was detected by immunohistochemical. Results:HE staining showed normal cell morphology structured in negative control group ; there were liquefied necrosis and cell swelling disorder in HIBD group ; EP group change was decreased compared with HIBD group. Immunohistochemistry showed that MMP--9 positive cells were decreased in the EP group compared with HIBD group but higher than the negative control group (P 〈 0. 01). Conclusion: ethyl pyruvate could play a protection role in HIBD by reduing the expression of MMP-9.
