摘要 目的探讨抑郁症患者MTHFR基因多态性与抑郁症发病的相关性。方法将94例抑郁症患者作为研究组,98名身心健康正常人为对照组,应用荧光偏振免疫法检测血清同型半胱氨酸水平,运用聚合酶链反应-限制性内切酶片段长度多态性分析技术(PCR—RFLP)检测MTHFR基因C677T多态性。入组时对研究组进行汉密尔顿抑郁量表(HAMD)评定。结果研究组与对照组基因型频率和等位基因频率分布差异有统计学意义(P<0.05)。研究组和对照组TT基因型Hcy浓度均较CT型、CC型高,且研究组TT基因型HAMD评分也比后两者高(P<0.05)。Logistic回归分析显示Hcy&基因型是抑郁症发病的危险因素。结论 TT基因型可能是抑郁症发病的重要危险因素之一,它通过影响血清同型半胱氨酸水平而影响抑郁症的严重程度。 Objective To investigate the relationship between C677T polymorphism of MTHFR gene with depression.Methods A total of 94 patients with depressive disorder in study group and 98 normal health controls in control group were involved in this study.The serum levels of homocysteine were determined by fluorescence polarization immunoassay(FPIA).Polymorphism of C677T was detected by using polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).Hamilton rating scale for depression-24 items(HAMD-24) was used to evaluate depressive symptoms.Results There were significant differences in the distribution of genotypic frequency and allele frequency between the study group and the control group(P0.05).The plasma Hcy levels in subjects with TT genotype in study group and control group were significantly higher than those with CT or CC genotype.Score of HAMD in patients with TT genotype in study group was significantly higher than those with other two genotypes(P0.05).Logistic regression analysis found that the Hcy genotype interaction was positively related with depression.Conclusion TT genotype of MTHFR gene may be the risk factor of the susceptibility of depression.It may affect the severity of depression by influenting the Hcy level.
