摘要
目的制备多孔纳米羟基磷灰石/壳聚糖(nHA/CS),对其进行细胞毒性检测。方法采用共沉淀法和粒子沥滤法制备多孔nHA/CS。分离培养大鼠成骨细胞,碱性磷酸酶(ALP)染色进行成骨细胞进行鉴定,倒置显微镜下观察培养过程中细胞的形态和增殖情况。MTT检测多孔nHA/CS支架材料对成骨细胞增殖的影响,评价细胞毒性。结果细胞可在多孔nHA/CS周围贴壁生长,并且附着、增殖,其生长及功能不受抑制。结论多孔nHA/CS无细胞毒性,有利于细胞的黏附和增殖,可以用于骨组织工程支架材料。
Objective To prepare porous nano-hydroxyapatite/chitosan scaffold(nHA/CS),and evaluate its cytotoxicity.Methods Porous-Nano-hydroxyapatite/chitosan were prepared through co-precipitation method and particle-leaching method.The rat osteoblasts were isolated and cultured.The cells were identified by cell Alkaline phosphatase(ALP) dyeing.And the morphdogy and proliferation of the cultured cells were observed under inverted microscope.The effects of porous-nHA/CS on the relative proliferation rate of rat osteoblasts were detected by MTT-assay.Results The cells adherently grew around the material,and the proliferation and growth were not affected by nHA/CS.Conclusion The porous-nHA/CS has no cytotoxicity in Vitro and also benefit to cell adhesion.The material can be used for bone tissue engineering.
