摘要
用有致痫作用的马桑内酯处理培养的大鼠皮层神经细胞。以原位DNA 断端标记法检测培养神经细胞的凋亡,配以培养液中乳酸脱氢酶的检测,观察马桑内酯对培养皮层神经细胞的毒性作用,并与已知的神经毒剂海人酸作比较。结果显示,马桑内酯可诱导培养大鼠皮层神经细胞凋亡,而引起细胞坏死的作用不如海人酸明显,其对培养大鼠皮层神经细胞的神经毒作用以导致神经细胞凋亡为主。
Apoptosis of cultured cortical neuronal cells of fetal rats was detected with in situ TUNEL assay, and colorimetric LDH assay was used to determine the neurotoxic activity of coriaria lactone(CL), a definitive epileptogenic agent, and compared with that of a known neurotoxicant kainic acid (KA). Results showed that CL can induce apoptosis on cultured cortical neuronal cells, but necrosis is not so obvious as compared to that of KA. This suggests that apoptosis is the major event involved in CL cytotoxicity in cultured cortical neuronal cell. (Figures 2~9 on plate 64)
