摘要
目的:探讨六味地黄丸对甲状腺功能亢进肾阴虚小鼠基础代谢的影响及其机制。方法:按每只小鼠每天灌胃3 mg四碘甲状腺原氨酸和20 mg利血平制备甲状腺功能亢进肾阴虚动物模型,连续给药21d。于造模当日开始分别灌胃给予不同药物,每日一次,连续给药21d。测给药前后体重、体温、心率和耗氧量。断头采血,取血浆用放射免疫分析法(RIA)测定环磷酸腺苷(cAMP);取血清用RIA测定三碘甲状腺原氨酸(T3)、四碘甲状腺原氨酸(T4)、游离三碘甲状腺原氨酸(FT3)和游离四碘甲状腺原氨酸(FT4)含量。结果:10g/kg、20g/kg和30g/kg六味地黄丸对体重、体温和心率均有明显作用;使存活时间延长,耗氧量降低;血浆cAMP水平下降,血清T3、T4、FT3、FT4含量下降。结论:六味地黄丸对甲状腺功能亢进肾阴虚证所致基础代谢增强有一定调节作用,减少机体的耗氧量。
Objective: To investigate the effects and mechanism of Liuwei Dihuang Wan on basal metabolism in Shenyin vacuity model mice with hyperthyroidism by thyroxin and reserpine.Methods: The Shen yin vacuity model mice were taken 3 mg thyroxin and 20 mg reserpine by administration by gavage every mouse every day once to prepare the model for 21 d,and at the same time Liuwei Dihuang Wan was taken at dose of 10 g/kg,20 g/kg and 30 g/kg by administration by gavage while model building.It was that determined body weight,temperature and heart rate from beginning to end and oxygen consumption to end.Plasma cAMP and triiodothyronie(T3),tetraiodothyronine(T4),free T3(FT3) and free T4(FT4) in serum were determined by radioimmunoassay(RIA).Results: Liuwei Dihuamg Wan could regulate changed the weight,temperature and heart rate(all P0.05,P0.01) at normal,prolong survival time(P0.01),reduce oxygen consumption,drop the level of plasma cAMP(P0.01) and the content of T3,T4,FT3,FT4 in serum(all P0.01) as Shen Yin vacuity model mice with hyperthyroidism and Shen Yin deficiency.Conclusion: It is demonstrated that Liuwei Dihuang Wan can reduce basal metabolism on hyperthyroidism and Shenyin deficiency,which maybe the key to it as vacuity-heat-clearing and Yin-supplementing.
