摘要
本文用高效液相色谱一电化学检测法,测定2例Joseph病患者脑脊液中多巴胺及其代谢产物含量变化,用内标法进行定量分析,以3.4-二羟基苄胺作为内标物;以5例非神经系统变性病为对照组。结果表明Joseph病患者脑脊液中DA、HVA、DOPAC的含量均比对照组低,P值分别为<0.05、<0.01、<0.05,有明显的统计学差异。此结果与文献报道相一致,为临床诊断提供了生化依据。文中对DA能系统的分布、降解代谢、功能以及与Joseph病的关系作了讨论。
The contents changes of cerebrospinal fluid (CSF) dopamine (DA) and metabolites were reported in 5 patients without nervous system degeneration as control group and 2 patients with Joseph disease. The DA and its metabolites (3, 4-Dihydroxyphenylacetic acid, DOPAC; homovanillice acid, HVA) were determinated by high-performance liquid chromatography with electrochemical detection (HPLC-EC), and were quantitated by internal standard method. 3, 4-dihydroxybenzamine (DHBA) was used as an internal standard. The results indicated that the DA, DOPAC and HVA levels were significantly lower in the Joseph disease group than those in the control. The contents in the Joseph disease group were 18. 97±5. 30, 51. 86± 10. 06 and 97. 86±9. 58 ng/ml respectively, while they were 2. 26± 0. 20, 5. 94 ±4. 94 and 31. 43 ±23. 96 ng/ml respectively in the controlgroup. The results agree with the references ever reported, and provided a biochemi cal basis of clinical diagnosis. The metabolism, distribution, function of DA and the relationship between DA and Joseph disease were also discussed.
