摘要
采用血小板聚集(PAG)试验比浊法观察发现穿心莲(APN)提取物 F_(01341)在一定剂量范围内呈剂量依赖性抑制二磷酸腺苷(ADP)和血小板活化因子(PAF)诱导的人富血小板血浆(PRP)的 PAG。其对 ADP 诱导第1min 和第5min PAG 的 IC_(50)分别为2.6mg/L和1.4mg/L;对由 PAF 诱导 PAG 的 IC_(50)分别为0.58mg/L 和0.38mg/L。血小板鲁米诺化学发光技术证明 APN F_(01341)2mg/L 即显著抑制静息及刺激后人洗涤血小板的化学发光。
Using a turbidimetric method,the authors found that Andrographis paniculate Nees extract F01341(APNF01341) dose-depandently inhibited platelet aggregation(PAG)induced by adenosine diphosphate(ADP)and platelet activating factor(PAF)in human platelet-rieh plasma(PRP).The IC50 values determined with a range of 0.6~4.8mg/L were,2. 6mg/L and 1.4mg/L for the first and second phase of PAG induced by ADP respectively.The IC50 values with dose range 0.3~1.2mg/L were 0.58mg/L and 0.38mg/L for the first and second phase of PAG induced by PAF respective- ly.APNF01341(2mg/L)also inhibited both resting and PMA or thrombin induced luminol-dependent chemiluminescence of human platelet in washed suspension.
