液相色谱-化学发光法测定丹参制剂中四种水溶性成分的含量 被引量:4

Simultaneous determination of four water-soluble components in salvia miltiorrhiza preparations by liquid chromatography with chemiluminescence detection

摘要 目的建立液相色谱-化学发光联用技术测定丹参制剂中四种水溶性成分(丹参素、原儿茶醛、丹酚酸B和丹酚酸A)含量的新方法。方法基于丹参四种水溶性成分对HAuCl4-luminol-H2O2化学发光的增强作用,用Shim pack ODS色谱柱(250mm×4.6 mm,I.D.5μm),以0.1%磷酸水溶液(v/v)和0.1%磷酸甲醇溶液(v/v)为流动相进行液相色谱梯度洗脱,与化学发光在线检测联用,在26 min内实现了四种丹参水溶性成分的高选择性和高灵敏分析。结果四种丹参水溶性成分的检测限在0.1~8.0μg.L-1的范围内,精密度(RSD)均小于2.5%,测定样品的回收率在95.3%到103.9%的范围内。结论本法选择性好、灵敏度高,重复性好,适用丹参制剂中水溶性成分的含量测定。 Aim To develop a novel method using high-performance liquid chromatography coupled with chemiluminescence detection for the simultaneous determination of four water-soluble components(danshensu,protocatechuic aldehyde,salvianolic acid B,and salvi-anolic acid A) in Salvia miltiorrhiza preparations.Methods Good separations of four water-soluble components were achieved on Shim-pack ODS column(250 mm × 4.6 mm,I.D.5 μm) with a gradient elution using the mixture of aqueous phosphoric acid(0.1%,v/v) and methanol phosphoric acid(0.1%,v/v) within 26 min.The detection limits(S/N = 3) for four water-soluble components were in the range from 0.1 to 8.0 μg·L^-1,respectively.Results The relative standard deviations(RSDs) of precision were below 2.5%.The mean recoveries ranged from 95.3% to 103.9%.Conclusion The proposed method is highly selective and sensitive,reproduci-ble,and suitable for the quality control of four water-soluble components in salvia miltiorrhiza preparations.
