黄芪、丹参对肥大心肌细胞SERCA2a、PLB mRNA表达的影响 被引量:9

Effect of radix astragali and Salvia miltiorrhiza on expressions of SERCA2a,PLB mRNA in hypertrophic cardiomyocytes

摘要 目的:研究黄芪注射液、丹参注射液对肥大心肌细胞SERCA2a、PLB mRNA的影响。方法:采用AngⅡ致乳鼠心肌细胞肥大模型,分为对照组、模型组、黄芪组、丹参组、联合组,采用RT-PCR法观察给药后24、48、72h心肌细胞PLB和SERCA2a mRNA水平。结果:AngⅡ可使PLB和SERCA2a mRNA水平降低,各时点SERCA2a下降均具有显著差异(P<0.05),48、72h PLB下降具有统计学意义(P<0.01),各用药组不同时点、不同程度改善PLB、SERCA2a mRNA水平与SERCA2a/PLB。结论:黄芪注射液、丹参注射液及其联合应用可以在不同时间改善SERCA2a、PLB mRNA水平和SERCA2a/PLB。 Objective:To study the effect of Raidx Astragali and Radix Salviae Miltiorrhizae on PLB and SERCA mRNA in cardiac hypertrophy and the mechanism of Raidx Astragali and Radix Salviae Miltiorrhizae against heart failure.Methods: The model of cardiac hypertrophy was induced by angiotersin Ⅱ.The culture cells were at random divided into six groups: control group,model group,Astragali group,Salviae group,combination treatment group.The effect of Raidx Astragali and Radix Salviae Miltiorrhizae in treatment on the level of PLB and SERCA2a mRNA transcription was observed by PT-PCR after administration for 24hour,48hour,72hour.Results:AngiotersinⅡ decrease the expression of PLB and SERCA2a mRNA.All the treated groups could increase the expression of SERCA2a mRNA and SERCA2a/PLB in different degree at different time.Conclusion: Raidx Astragali and Radix Salviae Miltiorrhizae in treatment could increase the level of SERCA and PLB mRNA transcription and improve the ratio of SERCA/PLB,and it might be one of the mechanism of Raidx Astragali and Radix Salviae Miltiorrhizae in against heart failure.
