摘要 目的:利用表面增强激光解析离子化飞行时间质谱仪(Surface-enhanced laser desorption-ionization time-of-flight mass spectrometry,SELDI-TOF-MS)及其配套蛋白质芯片筛选乳腺癌生物学标记物,初步探讨其在乳腺癌诊断中的临床意义。方法:用美国Ciphergen公司研制的蛋白指纹图谱仪和蛋白质芯片检测30例乳腺癌患者、30例乳腺纤维瘤患者、30例正常人血清蛋白质的相对含量,用SPSS 11.5软件分析处理数据,筛选标志物,建立诊断模型。结果:在乳腺癌、乳腺纤维瘤患者和正常人中共发现3个差异有统计学意义的蛋白质峰表达,质荷比(m/z)分别为:5700Da、8 900Da、5 910Da。用于乳腺癌诊断灵敏度和特异度分别为86.7%和96.7%。结论:蛋白指纹图谱技术在乳腺癌诊断及筛选生物学标记物等方面有很大的价值,值得进一步研究。 Objective: To screen the biomarkers of breast cancer by using the surface-enhanced laser desorption-ionization time-of-flight mass spectrometry (SELDI-TOF-MS) and protein chip array technology and to evaluate their clinical significance in the diagnosis of breast cancer. Methods: The serum proteomic patterns were detected in 30 patients with breast cancer, 30 patients with breast adenofibroma and 30 healthy blood donors. The diagnostic models were developed and validated by discriminant analysis. Resuits: The peak intensity of 5 700 Da,8 900 Da.5 910 Da (m/z) proteins were detected in the three groups of patients with significant difference. The diagnostic model made up of the differential expression proteins of 5 700 Da,8 900 Da,5 910 Da had a sensitivity of 86.7% and a specificity of 96.7%. Conclusion: The surface-enhanced laser desorption-ionization time-of-flight mass spectrometry (SELDI-TOF-MS) technique is of great value in the diagnosis and the biomarker screening of breast cancer and worthy of further study and wide application.
