摘要
目的 比较核酸提取仪和手工法核酸提取对荧光定量PCR准确定量的影响.方法 血清样本分别用NP968核酸提取仪和手工法提取DNA和RNA,提取产物作为模板,用ABI 7500进行荧光定量PCR分别检测HBV和HCV,对比CT值,比较两种核酸提取方法的效率.结果 核酸提取仪提取的核酸PCR扩增的CT值明显低于手工法的CT值,处理大批量样本的时间也明显短于手工法.结论 核酸提取仪具有速度快、提取效率高的优点,值得在临床实验室推广应用.
Objective To compare the nucleic acid extraction effect on accurate quantification in fluorescent quantitative PCR between hand-made method and nucleic acid extraction apparatus. Methods DNA and RNA of serum samples were extracted by NP968 nucleic acid extraction apparatus and hand-made method,respectively. The extracted products were used as fluorescent quantitative PCR template to detect HBV and HCV by ABI 7500. Results The CT values of nucleic acid extracted by apparatus was obviously lower than that of hand-made method. The samples handling time of was shorter than hand-made method, too. Conclusion Nucleic acid extraction using extraction apparatus is quick and efficient. It is worth to extend in clinical laboratory.
