胃肠道肿瘤患者门静脉血CK20mRNA和CEA mRNA检测及其意义

摘要 目的 检测胃肠道肿瘤患者门静脉血中CEA mRNA和CK-20 mRNA的表达.方法 采用荧光定量逆转录聚合酶链反应(FQ-RT-PCR)方法检测胃肠道肿瘤患者手术前后门静脉血中循环肿瘤细胞标志物CEAm RNA和CK20 mRNA的表达情况,并对上述指标在外周血中的含量进行检测;同时以非肿瘤手术患者作为对照组.结果 胃肠道肿瘤患者手术后门静脉血CK20 mRNA (79.7%)和CEA mRNA(84.4%)的阳性表达率与手术前(62.5%,67.2%)比较差异具有统计学意义(P〈0.05);手术前后门静脉血CK20 mRNA和CEA mRNA的阳性表达率与外周血(43.8%,51.6%)比较差异均具有统计学意义(P〈0.05).胃肠道肿瘤患者术后门静脉血中CK20 mRNA(mean=3.00)和CEA mRNA(mean=3.56)表达量与手术前(mean=2.34,2.58)和外周血(mean=1.16,1.93)比较差异均具有统计学意义(P〈0.05).结论 门静脉血中CK-20 mRNA,CEA mRNA的表达明显高于外周血. Objective To detect portal venous blood CK20 mRNA and CEA mRNA expression of patients with gastrointestinal cancer. Methods Using fluorescent quantitative reverse transcription polymerase chain reaction (FQ-RT-PCR) method to detection the tumor cells circulating markers CK20 mRNA and CEA mRNA expression in peripheral blood ,preoperative and post-operative portal venous blood with gastrointestinal cancer patients and non-tumor operation patients as a control group. Results The CK20 mRNA and CEA mRNA expression positive rates of post-operative portal venous blood with gastrointestinal cancer patients were 79.7% and 84.4% ;and higher than pre-Operative (62.5%, 67.2% )(P〈 0. 05). Both pre-operative and post-operative were significantly higher than peripheral blood (43.8%,51.6%) (P〈0.05). Experimental group and control group pre-operative and post-operatlve in portal vein and peripheral blood CK20 mRNA and CEA mRNA expression positive rate were significantly different (P^0. 001). The CK20 mRNA and CEA mRNA ex- pression values of post-operative portal venous blood with gastrointestinal cancer patients were significantly higher than both pre-operative portal vein and peripheral blood (P〈0.05). Conclusion CK20 mRNA and CEA mRNA were a high level in portal venous blood with gastrointestinal cancer patients more than peripheral blood.
