摘要
目的建立同时测定注射用头孢他啶(含精氨酸)中头孢他啶和精氨酸的含量的高效液相色普法(HPLC)。方法采用Thermo pHenyl-2柱,乙腈:醋酸盐缓冲液(5∶95)为流动相,检测波长206nm。结果头孢他啶、精氨酸线性范围分别为0.09196~0.9196mg/ml和0.04868~0.4868mg/ml,回收率分别为99.97%~100.59%和99.80%~101.59%,相对标准偏差(RSD)分别为0.04%~0.74%、0.02%~1.02%(n=9)。结论该法专属性好、快速、准确。
Objective To develop a reversed-phase high performance liquid chromatographic (RP-HPLC) method for the simultaneous determination of ceftazidime and arginine in injectable powder. Methods A HPLC method was established applying Thermo Hypersil Phenyl-2 Column (250 mm×4.6 mm 5μm), with a mobile phase of acetate buffer and acetonitrile (95:5). The detection wavelength is 206 nm with a flow rate at 1.0 ml/min and column temperature at 30℃. Results The linear ranges of ceftazidime and arginine were 0.091 96~0.919 6 mg/ml and 0.048 68~0.486 8 mg/ml, respectively. The average recoveries were 99.97%~100.59% for ceftazidime, and 99.80%~101.59% for arginine. The RSD values of inter-day and intra-day assays were 0.04%~0.74% and 0.02%~1.02% for ceftazidime and arginine, respectively. Conclusion The method developed is convenient, rapid, accurate and precise.
