摘要
目的研究萝藦多糖粗提物对免疫抑制小鼠免疫器官重量及淋巴细胞增值影响。方法将小鼠随机分为五组,分别是空白组、CY对照组、薄芝糖肽阳性对照组、多糖低剂量组、多糖高剂量组。多糖高、低剂量组(1 600 mg/kg、400 mg/kg)每天灌胃给药0.4 mL,阳性对照组腹腔注射给药(25 mg/kg)0.2 mL,空白组每天灌胃给药生理盐水0.2 mL,连续14 d。除空白对照组外,各组于给药第8天,腹腔注射环磷酰胺(25 mg/kg)0.1 mL/10g,连续7 d。末次给药24 h后,拉颈椎处死小鼠,称小鼠体重、脾、胸腺、肝脏重量,计算胸腺、脾指数。采用MTT法测定脾淋巴细胞的增殖情况。结果多糖高、低剂量组小鼠的体重及免疫器官指数高于CY对照组,脾淋巴细胞增殖均显著(P<0.05)高于空白组。结论萝藦多糖能提高免疫抑制小鼠体重及免疫器官指数,促进脾淋巴细胞增殖。
Objective To study the effects of the extracts of metaplexis japonica polysaccharides on the immune sup- pression of the weight of immune organs and lymphocyte proliferation of mice. Methods The mice were randomly divided into five groups, namely the blank group, CY control groups, bozhi glycopeptide positive control group, polysaccharides of low - dose group and high doses of polysaccharide group. Daily intragastric administration of polysaccharide high, low dose group ( 1 600 mg / kg, 400 mg/ kg) was 0.4 mL and intraperitoneal injection of Medicine Positive control group was (25 mg/ kg) 0. 2 mL which went on for 14 days. On the eighth day of medicine use, except those in the blank control group, each group were given intraperitoneal injection of cyclophosphamide (25 mg / kg) 0. 1 / 10 g, for 7 consecutive days. For the last time after administration for 24 h, the mice were killed by pulling the cervical and measured the weight of mice body, the spleen, thymus and liver then calculated the index of spleen, thymus gland. MTY method was applied to the measurement of splenic lymphocyte proliferation. Results The index of weight and immune organs of mice of poly- saccharide high group and low dose group was higher than that of CY group, the proliferation of splenic lymphocytes was significantly higher (P 〈 0.05) than that of the blank group. Conclusion Metaplexis japonica polysaccharides can im- prove the immune suppression of bodyweight and index of immune organs and promote the proliferation of splenic lympho- cytes of mice.
