摘要
目的:建立反相高效液相色谱法测定益抗灵合剂中芍药苷的含量。方法:采用反相高效液相色谱法,以Eclipse XDB-C18柱(4.6 mm×25 cm,5μm)分离样品,乙腈–0.1%磷酸溶液(15∶85)为流动相,流速1.0mL.min-1,检测波长230 nm,进样量10μL,柱温30℃。结果:在给定的色谱条件下,芍药苷与相邻组分峰之间分离良好,阴性对照试验无干扰。芍药苷浓度在19.1~95.7μg.mL-1之间与峰面积呈良好线性关系,回归方程Y=0.2415X+0.3207,r=0.9999,平均回收率(n=6)为103.4%;RSD=1.16%。结论:本法操作简便,结果准确,重现性好,可作为益抗灵合剂的含量控制方法。
Objective:To establish a method for determination of paeoniflorin in Yikangling Mixture by RP-HPLC.Methods:The RP-HPLC method was set up.The sample was separated and analyzed on a Eclipse XDB-C18 column(4.6mm×25cm,5μm),and acetonnitrile-0.1%phosphoric acid(15∶ 85) was used as mobile phase.The flow rate was at 1.0 mL·min-1.The detective wavelength was at 230 nm.Injection volume was 10 μL,column temperature was 30℃.Results:With this chromatographic condition,the paeoniflorin peak in Yikangling Mixture sample chromatogram could be separated with other ingredient peaks completely.The calibration curve was linear at a range of 19.1~95.7μg ·mL-1 for paeoniflorin with r =0.9999.The average recoveries(n=6) was 103.4%,RSD was 1.16%.Conclusion: This method is simple,accurate,reproducible,and used for quantitative control method of this preparation.
