摘要
目的观察腺苷A1受体激动剂对兔脊髓缺血/再灌注损伤的保护作用。方法新西兰大白兔随机分成治疗组、对照组和假手术组,建立兔脊髓缺血/再灌注模型,给予腺苷A1受体激动剂CPA、DMSO,观察兔神经功能恢复;通过病理切片,计算脊髓前角正常神经元;应用免疫荧光染色检测Bcl-2表达。结果治疗组和假手术组的神经功能评分在各观察时间点均明显高于对照组(P<0.01);与对照组相比,治疗组和假手术组的脊髓前角正常神经细胞数明显增多(P<0.01);治疗组兔脊髓神经元的免疫荧光反应明显强于对照组。结论腺苷A1受体激动剂对脊髓缺血/再灌注损伤具有保护作用,可保护神经元和改善神经功能障碍。
Objective To investigate protective effect of adenosine A1 receptor agonist on spinal cord ischemia/reperfusion injury.Methods New Zealand rabbits were divided into three groups randomly: treatment group,control group and sham-operated group.Adenosine A1 receptor agonist,CPA,was applied to spinal cord ischemia/reperfusion injury rabbit model.Then the recovery of neuron functions was evaluated and the numbers of normal neurons in anterior horn of spinal cord were calculated.The Bcl-2 expression was detected by immunofluorescent staining.Results Scores of neurologic functions in treatment and Sham group were higher than that in control group(P0.01).More normal neurons in anterior horn of spinal cord were found in CPA group compared with control group(P0.01).Conclusion Adenosine A1 receptor agonist can protect spinal cord ischemia/reperfusion by saving normal neurons and rescuing neuron functions.
