摘要
目的获取野生与栽培茅苍术差异遗传信息。方法运用RAPD分子标记方法,从DNA水平上分析野生与栽培茅苍术的遗传变异。结果筛选了60个随机引物,从中选取18个多态性强、重复性好的引物进行扩增,共检测到228个位点,110个多态性位点,多态位点比率为48.25%;野生与栽培茅苍术的遗传距离为0.32。结论野生和栽培茅苍术基因组DNA存在显著差异,研究工作为茅苍术优良种质资源鉴定和可持续利用提供科学依据。
OBJECTIVE To assess the genetic variation between wild and cultivated Atractylodes lancea(Thunb.) DC.METHODS Genetic diversities between wild and cultivated Atractylodes lancea(Thunb.) DC.at DNA level were analyzed by using RAPD marker methods.RESULTS 60 random primers were screened,out of which 18 primers with high polymorphism and good repeatability were chosen to amplify.As a result,228 loci were detected.Among them,118 loci were polymorphic,presenting a polymorphic rate of 48.25%.The genetic distance between wild and cultivated Atractylodes lancea(Thunb.) DC.was 0.32.CONCLUSION There were significant differences in genome DNA between wild and cultivated Atractylodes lancea(Thunb.) DC.This work provides a foundation for identifying high quality germplasm resource and sustainable utilization of Atractylodes lancea(Thunb.) DC.
