局部注射不同浓度骨保护素对大鼠正畸牙移动影响的研究 被引量:6

摘要 目的:研究局部注射不同浓度骨保护素对大鼠牙齿移动的影响。方法:选用10周龄雄性SD大鼠60只,分为实验组与对照组,实验组分0.5mg/kg OPG组和0.05mg/kg OPG组,每组20只。在右上颌第一磨牙施加40G左右力使其移向近中。在实验组和对照组大鼠右上颌第一磨牙腭侧粘骨膜下分别注入重组人骨保护素rhOPG-Fc和生理盐水。在ld、4d、7d、14d分别处死实验组与对照组大鼠各5只,通过模型测量实验组与对照组右上颌第一磨牙的移动距离,HE染色观察右第一磨牙近中根压力侧牙周组织形态的变化和TRAP染色观察破骨细胞数量的变化。结果:1.在7天和14天时注射OPG的两组大鼠的牙齿移动量显著低于对照组,同时高浓度0.5mg/kg OPG组的移动量显著小于低浓度0.05mg/kg OPG组。2.对照组压力侧能看到显著的牙槽骨吸收,而注射OPG组大鼠牙槽骨吸收不明显。3.TRAP染色发现4天、7天和14天时第一磨牙近中根压力侧破骨细胞数量两个实验组显著少于对照组,而0.5mg/kg OPG组破骨细胞数量显著少于另外两组。结论:局部注射OPG后能显著减少破骨细胞的产生,同时能显著降低牙齿移动距离。高浓度的OPG比低浓度的OPG对抑制牙齿移动的作用更大。 Objective: To approach the effects of local dilivery of different kind of OPG to tooth movement in rats. Method: Sixty ten - mouths old male Spargue- Dewley rats were divided into three groups which were control group, 0.5mg/kg OPG group and 0.05mg/kg OPG groap. Each group contains 20 rats. About 40g force loaded in the right first molar in order to pull it mesio. Humens' ostecprotegerin and physiological saline were injected into the palatine- side' s mucoperiosteum of the right upper first molar of the experimental group and the control group respectively. Five rats of each experiment group and control group were sacrificed in I d, 4d, 7d and 14d .To measure the movement of the right first molar by the model. The change of the periodental form and the quantity of the osteoclast were observed by histological section and TRAP staining. Result:1. The moving distance of the right upper first molar of the two experimental groups in 7d and 14d is apparently lower than that of the control group ( p 〈 0. 001 ). And the moving distance of the right upper first molar of the 0.5mg/kg OPG group is lower than the 0.05mg/kg OPG group. 2. The remarkable resorption of the alveolar bone can be seen in the control group. 3. The quantity of osteoclasts in the pressure side of the mistal root of the right first upper molar in the two experimental groups varied in 4d, 7d and 14d was obviously less than that in the control group (p 〈 0. 001 ). And the quantity in the 0.5mg/kg OPG group is less than two other groups. Conclusions: Local delivery of OPG inhibits osteoclastogenesis and tooth movement. And the high dose OPG is more remarkable.
