消栓通脉颗粒对深静脉血栓形成大鼠静脉壁NF-κB,IκB表达的影响 被引量:3

Effect of Xiaoshuan Tongmai Granules on Level of NF-κB and IκB in Vein Wall in Rats with in Deep Venous Thrombosis

摘要 目的:探讨中药消栓通脉颗粒治疗深静脉血栓形成的机制。方法:72只Wistar大鼠随机分为4组:假手术组、血栓模型组、复方丹参片组(0.23 g.kg-1,ig)、消栓通脉颗粒组(32 g.kg-1,ig)。连续给药5 d后采用下腔静脉结扎法制备血栓模型,造模后4 h连续给药,动态监测各组大鼠术后不同时间点静脉壁核因子κB mRNA(NF-κB p50 mRNA,NF-κB p65mRNA),抑制因子κB mRNA(IκBαmRNA)表达水平。结果:同一时间点,中药治疗组静脉壁NF-κB p50 mRNA,NF-κB p65mRNA表达水平显著低于模型组(P<0.01),而IκBαmRNA明显增高(P<0.01);各组静脉壁NF-κB p50 mRNA,NF-κB p65mRNA表达随手术天数的延长呈现先增强后减弱的趋势,术后3 d达到高峰,而IκBαmRNA的表达呈先下降后升高的规律,术后3 d降到低谷;与复方丹参片组比较,在同一时间点,消栓通脉颗粒剂组NF-κB p50 mRNA,NF-κB p65 mRNA表达明显降低(P<0.01),而IκBαmRNA表达显著增高(P<0.01)。结论:消栓通脉颗粒剂可能通过调控NF-κB/IκB信号传导通路,减轻其介导的静脉壁炎症反应,保护血管内皮细胞功能,发挥抗血栓作用。 Objective: To explore the interventional mechanism of Chinese herbs Xiaoshuan Tongmai Granules(XSTM) on deep venous thrombosis(DVT)in rats.Method: Seventy two Wistar rats were randomly divided into 4 groups: sham group,DVT model group,XSTM group,Fufang Danshen tablets(DS) group.Venous thrombosis was induced by inferior vena caval occlusion by ligature just below the level of the renal veins.The levels of NF-κB p50 mRNA,NF-κB p65 mRNA and IκBα mRNA in vein wall of all rats were dynamic detected at 1,3,6 days after model built.Result: At the same time,the levels of NF-κB p50 mRNA and NF-κB p65 mRNA in Chinese herbs treatment groups were significant lower than those in model group(P0.01),while the level of IκBα mRNA was significantly higher(P0.01).The levels of NF-κB p50 mRNA and NF-κB p65 mRNA were increased the decreasing trend after model induced,and reached the peak at 3rd day after ligature(P0.01) in all groups.But the levels of IκBα mRNA were decreased the increasing trend after model induced,and reached the low at 3rd day after ligature(P0.01) in all groups.At the same time,the levels of NF-κB p50 mRNA and NF-κB p65 mRNA in XSTM group were significantly lower than those in DS group(P0.01),while the level of IκBα mRNA in XSTM group was significantly higher(P0.01).Conclusion: It suggested that XSTM could intervene the system of NF-κB p / IκB to supress inflammatory reaction and thrombosis.
