摘要
分别采用TRIzol法和改良的CTAB法提取阔叶薰衣草叶片的RNA,并用紫外光谱和琼脂糖凝胶电泳对其提取效果进行鉴定。实验结果表明:TRIzol法提取的总RNA浓度高,但杂质较多且有部分降解、稳定性差;改良的CTAB法提取得到的总RNA的纯度较高,OD260/OD280值为1.909,效果稳定,有良好的可重复性。因此,改良的CTAB法是一种较为简便、经济的提取阔叶薰衣草叶片总RNA的方法。
In this research,TRIzol method and modified CTAB method to extract total RNA from leaves of Lavandula spica were employed,and some identification on its extracting efficiency by using ultraviolet spectrum and agarose gel electrophoresis were made.It indicated that the total RNA extracted by TRIzol was at high concentrations with a lot of impurities and some was degraded with poor stability.The purity of RNA isolated by using modified CTAB method was higher than TRIzol,with OD260/OD280 value of 1.909,which has stable performance and good repeatability.Therefore,the modified CTAB method is relatively simple and economical method for the RNA isolated from leaves of Lavandula spica.
