摘要 目的观察mPEG-SPA(甲氧基聚乙二醇-琥珀酰亚胺丙酸酸酯)对人红细胞RhD血型抗原修饰的效果,并探讨mPEG-SPA修饰的可行性。方法采用mPEG-SPA修饰人红细胞血型RhD抗原,并观察其对红细胞形态、结构和功能的影响。结果 2.0 mmol/L的mPEG-SPA在pH8.0室温条件下可有效遮蔽红细胞表面RhD抗原,且与红细胞结合牢固,对红细胞的形态、渗透脆性、自身溶血率、红细胞膜胆固醇含量、乙酰胆碱酯酶活性、Na+-K+ATP酶活性、2,3-DPG含量、血沉值、变形指数和常规指标均无明显影响。结论采用2.0 mmol/L mPEG-SPA遮蔽人红细胞RhD抗原获得了初步成功。 To discuss the feasibility of mPEG-SPA modification,we observed the effects of mPEG-SPA modification on blood group RhD antigen on human red blood cells(RBCs).mPEG-SPA was used to modify blood group RhD antigen on human RBCs,and subsequently the effects on morphology,structures,and functions of human RBCs was observed.mPEG-SPA could conjugated with RhD antigen firmly and camouflage RhD antigen efficiently when the concentration reached to 2.0 mmol/L at room temperature at pH8.0.mPEG modification did not demonstrate obvious effects on RBCs' morphology,osmotic fragility,hemolysis,cholesterol contents,AchE activity,Na+-K+ ATPase activity,2,3-DPG contents,ESR,deformability or routine index.All thses results suggest that camouflaging blood group RhD antigen on human by mPEG-SPA is preliminarily achieved.
