摘要 目的:体外培养的成纤维细胞(Fbs)与自体毛发角蛋白(HK)颗粒进行复合培养,探讨HK对Fbs的作用。方法:实验分为Fbs组、Fbs∶HK=1∶1组、Fbs∶HK=2∶1组及HK组。利用消化分离法对人Fbs进行原代培养;MTT法测定Fbs的生长曲线以及利用羟脯氨酸(HYP)试剂盒对Fbs与HK复合培养的培养液中HYP含量进行测定。结果:人皮肤Fbs生长状态良好;MTT实验表明,在Fbs与Hk的比例为2∶1时效果最为明显。在Fbs与HK的比例为2∶1时,培养液中的HYP含量最多。结论:体外培养的Fbs与Hk粒复合,可明显促进细胞分裂增殖的功能;在二者比例为2∶1时有利于促进Fbs合成胶原蛋白。 Objective To explore the effect of autologous hair keratin(HK) composite particles on fibroblasts(HK) in vitro.Methods The experiment was divided into Fbs group,Fbs∶HK=1∶1 group,Fbs∶HK=2∶1 group and HK group.The digestion method was used in primary culture of Fbs;MTT method was used to determine the growth curve of Fbs;the hydroxyproline(HYP) content in composite culture of Fbs and autologous HK was determined with HYP kit.Results The human skin Fbs grew well.When the ratio of Fbs to HK was 2∶1,the HYP content was the highest in composite culture.In MTT assay,when the radio of Fbs to HK was 2∶1,the grafting efficiency was perfect.Conclusion Composition of cultured Fbs in vitro with HK can significantly promote the fragmentation and proliferation of cells;when the ratio of Fbs to HK is 2∶1,it is benifit to promote the synthesis of Fbs into collagen.
