杀虫安对小鼠脑组织内神经递质的影响

摘要 研究了杀虫安经灌胃给药后对小鼠脑组织内多巴胺及氨基酸含量的影响。将60只ICR(美国癌症研究所)小鼠随机分为空白对照、杀虫安低剂量(200 mg/kg bw)及高剂量(300 mg/kg bw)3个处理组,于灌胃后出现惊厥时断头取脑,采用高效液相色谱-电化学(HPLC-ECD)检测法测定了鼠脑组织内多巴胺(DA)及其代谢产物二羟苯乙酸(DOPAC)和高香草酸(HVA)的含量,采用液相色谱-质谱联用(LC-MS/MS)法测定了氨基酸的含量。结果发现,两个剂量处理组鼠脑组织内DA及氨基酸含量与对照相比均有显著性差异。其中:DA比对照分别升高25.25%和31.93%(P<0.05),DOPAC分别升高23.16%和37.71%(P<0.05),天门冬氨酸(Asp)分别升高13.65%和15.07%(P<0.05),谷氨酸(Glu)分别升高20.07%和25.66%(P<0.05);而低剂量组γ-氨基丁酸(GABA)含量比对照降低32.55%(P<0.05),两个剂量处理组甘氨酸(Gly)含量分别比对照降低了13.26%和17.79%(P<0.05)。表明杀虫安导致惊厥时伴有鼠脑内多巴胺和氨基酸含量的变化。 The effects of profurite-aminium on the contents of dopamine and amino acids in mice brain by intragastric administration were studied.Sixty ICR(Institute of cancer research) mice were randomly divided into control group,low dose group(200 mg/kg bw)and high dose group(300 mg/kg bw) of profurite-aminium.The mouse brain was taken out after the mice were induced convulsion by intragastric administration,the contents of dopamine and it's metabolites(DOPAC and HVA) in brain of mice were determined by HPLC-ECD,and the contents of amino acids were determined by LC-MS/MS.The results showed that compared with control group,the low and high dose groups had significant difference in the contents of dopamine and amino acids in brain of mice.Among them,the contents of dopamine were increased 25.25% and 31.93%(P0.05) against the control group,the contents of dihydroxyphenylacetic acid were increased 23.16% and 37.71%(P0.05),the contents of aspartic were increased 13.65% and 15.07%(P0.05),the contents of glutamate were increased 20.07% and 25.66%(P0.05),respectively.While the contents of GABA of the low dose group were decreased 32.55%(P0.05) against the control group,the contents of glycine of the low and high dose groups were decreased 13.26% and 17.79%(P0.05),respectively.The results showed that contents of dopamine and amino acids in brain of mice were changed when the convulsion was caused by profurite-aminium.
