摘要 通过构建NMH7基因与绿色荧光蛋白基因融合的植物表达载体GFP-NMH7研究紫花苜蓿基因编码蛋白的亚细胞定位。用基因枪轰击洋葱表皮细胞的方法进行瞬时表达,结果表明:NMH7基因编码蛋白定位于细胞核;半定量RT-PCR分析紫花苜蓿NMH7基因的组织表达特性,结果表明:其在根、花蕾、花、荚果中都有表达,在花中表达量高于其他组织;NMH7基因的表达不受赤霉素诱导,脱落酸和茉莉酸甲酯能抑制其表达。 The subcellular location of gene NMH7 in alfalfa was studied,a recombinant plasmid with a green fluorescent protein gene(GFP NMHT) was constructed and transformed into onion epidermis cells by gene gun method. Transient expression analysis indicated that GFP-NMH7 fusion protein was localized in the nucleus of onion epidermal cells. Semi quantitative RT-PCR analysis was used to determine the tis- sue expression patterns of gene NMH7 in alfalfa, the results revealed that it expressed itself in roots, flo ral buds, flowers and pods, and there was the highest expression level in flowers. The results of Semi- quantitative RT-PCR analysis showed that the expression of gene NMH7 in alfalfa was not induced by gib- berellin and the expression of NMH7 gene in alfalfa was inhibited by Jasmonic acid methyl ester.
