摘要
目的:建立大鼠Ⅱ型糖尿病模型,检测并比较糖尿病大鼠种植体周围骨组织的整合素α5β1和纤维粘连蛋白(fibronectin,FN)表达水平,探讨Ⅱ型糖尿病影响种植体骨结合的可能机制。方法:建立糖尿病大鼠实验动物模型成功后,将48只大鼠随机分为正常种植组(C组)、正常对照组(C0组)、糖尿病种植组(T组)、糖尿病模型组(T0组)。在T组及C组大鼠右侧胫骨近骺端种植纯钛种植体,每组于植入2、4、8周分别处死4只大鼠,采用免疫组化方法检测各组种植体周围骨组织整合素α5β1和FN表达水平。结果:种植术后2周和4周时,T组种植体周围整合素α5β1和FN的表达水平明显高于C组,差异有统计学意义(P<0.05)。术后8周,T组与C组间整合素α5β1的表达水平接近,而T组FN的表达水平显著高于C组(P<0.05)。结论:糖尿病可能通过改变种植体周围组织中整合素α5β1及FN表达,削弱了种植体骨结合。
Objective：To detect and compare the expression of integrin α5β1 and fibronectin（FN） in osseous tissue around implant between Type Ⅱ Diabetic Rats and normal rats,and explore the possible mechanism that diabetic mellitus affect the implant osseointegration.Method：Establish the rat model of type Ⅱdiabetes,then 48 rats were divided into four groups randomly：normal implanting group（group C）,normal control group（group C0）,diabetic implanting groups（group T）,diabetic control group（group T0）.The group C and group T were implanted the titanium implant near the epiphyseal end of tibia.Rats were killed after two,four and eight weeks in batches.Using immunohistochemistry method the expression of integrin α5β1 and FN were detected in osseous tissue around the implants in each group.Result：The expression of integrin α5β1 and FN in osseous tissue around the implants in group T were significantly stronger than the expression in group C in two and four weeks（P 0.05）.Eight weeks after implantation,the expression of integrin α5β1 in group T were close to that of group C,while the expression of FN in group T was significantly stronger than group C（P 0.05）.Conclusion：Diabetic mellitus may weaken the implant osseointegration by changing expression of integrin α5β1 and FN in osseous tissue around the implants.
