摘要
对影响慈竹硫酸盐浆纤维素酶水解的主要影响因素作了探讨,包括pH、温度、液比、酶用量、时间、打浆度和吐温80用量;还对吐温80影响酶水解的原因进行了探讨。结果表明:在pH值4.8、温度45℃、底物质量浓度28.6g/L、酶用量25FPU/g、吐温80用量1mg/g的条件下水解52h,打浆度13°SR的浆料酶水解得率为82.39%。吐温80用量对慈竹硫酸盐浆酶水解的影响表现为:一方面影响酶的活性,另外一方面降低纤维素酶与木素的无效吸附,使游离酶的数量增加。
The main factors of influencing Sinocalamus affinis enzyme hydrolysis were investigated, which mainly include pH, reaction tem- perature, the solid-liquid ratio, the content of enzyme, the reaction time, the beating degree and the content of Tween 80. Besides, the hydrolysis mechanism of Tween 80 was also studied in this paper. The results show that the hydrolysis yield of 13°SR pulp is 82.39% at the conditions of pH 4.8, reaction temperature 45℃, substrate concentration 28.6g/L, enzyme content 25FPU/g, Tween 80 lmg/g and reaction time 52h. The content of Tween 80 can not only affect the activity of the enzyme, but also affect the adsorption and increase the free enzyme.
