比较基因组杂交芯片筛选头颈部鳞状细胞癌患者转移相关的差异表达基因 被引量:2

Differential expression of metastasis-related genes in patients with head and neck squamous cell carcinoma:a genomic hybridization microarray study

摘要 目的检测头颈部鳞状细胞癌(head and neck squamous cell carcinoma,HNSCC)标本,寻找与HNSCC转移有高度相关性的基因。方法采用比较基因组杂交芯片技术,分别检测80例临床HNSCC标本,包括舌癌17例,口腔癌13例,口咽癌14例,下咽癌36例。结果比较基因组杂交CGH检测结果示同未发生转移的HNSCC组织标本相比,发生转移的HNSCC组织标本内,基因扩增有显著性差异的位置有1q、4q、7q、11q、Xq,基因缺失有显著性差异的位置有5p、8p、9p、11q、13q、17p。Cox单因素分析发现这些差异基因主要涉及到转录调控和信号传导等功能分类。结论 HNSCC标本中存在与HNSCC转移相关的扩增基因和缺失基因。 Objective To detect the metastasis-related genes in patients with head and neck squamous cell carcinoma(HNSCC).Methods HNSCC metastasis-related genes were detected with genomic hybridization microarray in 80 samples from HNSCC patients,including 17 from tongue cancer patients,13 from oral cancer patients,14 from oropharynx cancer patients,and 36 from hypopharynx cancer patients.Results Genomic hybridization CGH showed that significant differences were observed in gene amplification at the sites of 1q,4q,7q,11q and Xq,and in gene deletion at the sites of 5p,8p,9p,11q,13q and 17p between the samples with and without HNSCC metastasis.Cox univariate analysis showed that the differentially expressed genes were mainly related with the functions of transcription regulation and signal transduction,etc.Conclusion HNSCC metastasis-related gene amplification and deletion can be found in samples from HNSCC patients.
