摘要
目的探讨麻黄加术汤对类风湿性关节炎的作用机制。方法运用Ⅱ型胶原法建立Wistar大鼠类风湿性关节炎模型,采用麻黄加术汤给予干预,以甲氨喋呤片为阳性对照药。将大鼠随机分为空白组、模型组、甲氨喋呤组、麻黄加术汤组。检测大鼠血清炎性细胞因子IL-1β、TNF-α浓度,观察大鼠膝关节滑膜组织病理改变。结果麻黄加术汤可显著降低大鼠血清炎性细胞因子IL-1β、TNF-α的含量,差异具有统计学意义(P<0.01),对CIA大鼠滑膜组织的炎性细胞浸润、纤维组织增生和巨噬样A型细胞有明显抑制作用,差异具有统计学意义(P<0.01)。结论麻黄加术汤治疗类风湿性关节炎的机制可能与降低炎性因子IL-1β、TNF-α的水平,抑制炎性细胞浸润、纤维组织增生和巨噬样A型细胞增生有关。
Objective To investigate the mechanism of Mahuang Jiazhu decoction on rheumatoid arthritis. Methods The arthritis models of Wistar rats were stablished by the type II collagen, the decoction was given to be therapeutic drug, and methotrexate tablets was used as a positive control The rats were randomly divided into control group, model group, methotrexate group, the decoction group. The serum concentration of inflammatory cytokines IL-1β TNF-α, and the synovial tissue pathology in knee joint was measured. Results The decoction significantly reduced the serum levels of inflammatory cytokines IL-1β and TNF-α content of synovial (P〈0.01), and inhibited significantly the CIA infiltration of inflammatory cells, fibrous tissue hyperplasia and macrophage-like cells-A significantly (P〈0.01). Conclusion The mechanism of the decoction for rheumatoid arthritis maybe related to reduce the inflammatory factor IL-1β and TNF-α levels, inhibit the inflammatory cells infiltration, fibrous tissue hyperplasia and maerophage-like cell proliferation-A.
