摘要
采用镰孢菌选择性培养基(FS)、PDA分离培养基和SNA鉴定培养基,针对对虾养殖环境中水域、沉泥及饲料等样本进行镰孢菌的分离,再用芽孢杆菌抗菌肽作用于分离株,观察其抑菌效应。结果共分离到镰孢菌255株,鉴定出14株梨孢镰孢菌和16株串珠镰孢菌。定量计算出水样镰孢菌平均浓度16 CFU/mL,沉泥样镰孢菌平均浓度161 CFU/mL,饲料样镰孢菌平均浓度为69 CFU/g。抑菌试验表明芽孢杆菌抗菌肽对受试菌株的气生菌丝均有抑制效应,最佳稀释度为2.5μg/mL。对虾养殖环境中存在一定浓度的镰孢菌,对对虾养殖存在潜在威胁;芽孢杆菌抗菌肽对对虾养殖环境中镰孢菌的抑菌效应显著。
The Fusarium species were isolated and identified by Fusarium selective medium(FS)isolation medium(PDA)and identifying medium(SNA)from the water,soil and feedstuff of shrimp culture environment.Then the inhibitory effect of antimicrobial peptide to isolates were observed.Results total 255 strains of Fusarium,of which 14 identified as F.poae and 16 as F.moniliforme was obtained.Quantitative calculation showed that mean concentration of Fusarium in water,soil and feedstuff were 16 CFU/mL,161 CFU/mL and 69 CFU/g.Bacteriostatic test indicates antimicrobial peptide could inhibit the growth of isolates,the best inhibitory concentration was 2.5 μg/mL.There is a certain concentration of Fusarium in shrimp culture environment,it may have potential threat to the growth of shrimp.Antimicrobial peptide's inhibitory effect to isolates is obvious.
