培养基对解淀粉芽孢杆菌ES-2菌株产抗菌脂肽的影响 被引量:47

摘要 【目的】研究培养基对解淀粉芽孢杆菌ES-2菌株产生抗菌脂肽的影响,为选择合适的产抗菌脂肽的培养基提供指导。【方法】通过HPLC技术对培养基中的碳、氮源对ES-2菌株抗菌脂肽组分的影响进行试验。【结果】改进的Landy培养基抗菌脂肽产量较高;葡萄糖是ES-2菌株产生脂肽的良好碳源,特别是对surfactin的产生有明显的促进作用;蔗糖和乳糖也是产生fengycin的良好碳源;L-谷氨酸和L-天门冬酰胺是抗菌脂肽产生的良好氮源;而不同的氨基酸组合并不有利于抗菌脂肽的产生;实验结果还显示,6种脂肪酸对抗菌脂肽的产生起到抑制作用。【结论】改进的Landy合成培养基是ES-2菌株产生脂肽适宜的培养基,在抗菌脂肽生产中向培养基中加入合适种类的糖和氨基酸十分重要。 [ Objective] In order to screen the medium for antimicrobial lipopeptide production, the influence of medium on the production was discussed. [ Method] Influence of carbon and nitrogen source of medium on antimicrobial lipopeptide component was examined by HPLC. [Result] The medium screening experiment indicated that the ES-2 could obtain high yield of antimicrobial peptides in the Landy medium. The result showed that glucose is a good carbon source for production of fengycin and surfactin, especially, which obviously promoted surfactin production; the sucrose and the lactose also were good carbon sources for production of surfactin. Therefore, different sugars in medium have certain influences on production of the two kinds of lipopeptide composition. The L-glutanic acid and the L-asparagine are good nitrogen sources for production of the antimicrobial lipopeptide. But the combination of different amino acids is certainly disadvantageous to the antimicrobial lipopeptide production. The experimental result also showed that 6 kinds of fatty acids inhibited antimicrobial lipopeptide production. [Conclusion] One kind of improved Landy synthesis medium is determined and the appropriate sorts of sugar and amino acid in medium components are extremely important for the antimicrobial lipopeptide production.
